Methods TCGA and geo database served as supporters to provide us with clinical sample data. Kaplan Meier method was used for overall survival analysis. The expression level of linc00511 was detected by 5 cell lines. CCK-8 cell counting Kit (CCK-8), colony forming assay and Transwell assay were used to detect the effect of linc00511 on cell behavior. The correlation between linc00511 and miR-195 or Eya1 was confirmed by luciferase reporter gene detection. The mRNA and protein expression levels were detected by quantitative real-time PCR and Western blotting. This study found that linc00511 was highly expressed in HCC tissue samples and cell lines, which may be related to poor prognosis and clinical parameters of HCC patients. Loss of function assay showed that linc00511 deletion inhibited the proliferation, colony formation and invasion of HepG2 cells, while functional gain assay showed that Huh7 cells had antagonistic effect. Bioinformatics tools and luciferase reporter gene detection showed that linc00511 may be a competitive endogenous RNA (Cerna) of miR-195, which was positively correlated with Eya1, and rescue experiment strengthened this role. In conclusion, linc00511 interacts with Eya1 to promote the occurrence and development of HCC by mediating miR-195, which provides a promising therapeutic marker for the diagnosis and prognosis of HCC.
