论文题目:Antibiotic-Pressure Enrichment of Anti-Mycobacterium tuberculosis Strains from Taklamakan Desert Soil
摘要:Drug-resistant tuberculosis continues to demand microbial resources that are not only taxonomically diverse but also biologically biased toward antimycobacterial chemistry. Desert soils provide such a setting because resident actinomycetes are shaped by chronic drought, oligotrophy, irradiation, and chemical competition. Here, we used rifampicin, streptomycin, and kanamycin as antitubercular selective pressures to enrich Taklamakan Desert soil communities in situ and to test whether this ecological filtering could improve the recovery of anti-Mycobacterium tuberculosis H37Ra-active actinomycetes. Twenty-one soil samples were exposed to each antibiotic at a final concentration of 1% and incubated in the original desert habitat for one month. Amplicon sequencing showed that antibiotic pressure reassembled the bacterial community rather than simply reducing diversity, with clear increases in actinobacterial representation, particularly after streptomycin treatment. Subsequent cultivation yielded 270 actinomycete isolates assigned to 7 orders, 8 families, 10 genera, and 79 species. Streptomycin-treated soils produced the largest isolate set, while Gause's No. 1 medium gave the highest recovery efficiency. Primary Alamar Blue screening identified seven strains with anti-M. tuberculosis H37Ra activity: Streptomyces atrovirens TRM 53043, S. nitrosporeus TRM 53047, S. amritsarensis TRM 53093, S. olivaceus TRM 53094, S. pimonensis TRM 53204, Actinomadura deserti TRM 53223, and S. marokkonensis TRM 53262. Draft-genome mining showed that these active isolates carried resistance-associated annotations, mobile-element-related genomic contexts, and diverse biosynthetic gene cluster repertoires. Together, the data support a working model in which antitubercular antibiotic pressure enriches tolerant, metabolically equipped actinomycetes, thereby increasing the practical likelihood of recovering strains with antimycobacterial activity for downstream natural product discovery.
